The Primer Dimer Effect on Real Time and qPCR
Primer dimers will only be detected in the amplification plot when an intercalating agent is used to detect amplification. However, even when a probe is used for detection (eliminating all mispriming events from the amplification plot) primer dimer formation causes a number of problems. Since their formation is not reproducible from well to well, quantification is less accurate and duplicate reactions on the same plate often donot pair well. Dimer formation also - Use up limited PCR reagents (primers and dNTPs) Reduce the priming efficency on target temp late Reduces the dynamic range
